چکیده

The synergistic antitumor action of 6-mercaptopurine (6-MP) and 6-methylmercaptopurine ribonucleoside (MMPR) is confirmed with Sarcoma 180 ascites cells. Striking decreases are seen in the adenine nucleotide concentrations in cells incubated for 1 hr with this drug combination, and lesser effects are seen on the guanine nucleotide concentrations. Still greater decreases in adenine nucleotide levels are observed when the cells are preincubated for 30 min with MMPR before addition of 6-MP. The 5'-monophosphate nucleotides of 6-MP and MMPR

Reversal of methylmercaptopurine ribonucleoside cytotoxicity by purine ribonucleosides and adenine.

6-Methylmercaptopurine ribonucleoside-5'-phosphate (MeSPuRMP), the sole metabolite of 6-methylmercaptopurine ribonucleoside (MeSPuRib), is a strong inhibitor of purine de novo synthesis, inducing depletion of intracellular purine nucleotides and subsequent cell death in several tumor cell lines. In this study prevention of MeSPuRib cytotoxicity by compounds of the purine salvage pathway was stu...

Biochemical Mechanisms for the Synergism between 6-Thioguanine and 6-(Methylmercapto)purine Ribonucleoside in Sarcoma 180 Cells1

of MMPR forms rapidly to levels of 1 to 2 mM in the tumor cells, and the steady-state levels of 5-phosphoribosyl 1-pyrophosphate increase 4to 5-fold in 6 to 12 hr. This permits a significantly greater synthesis of 6-thioguanosine monophosphate (6-thioGMP) after injection of 6-thioguanine. These findings are similar to those in previous reports concerning the synergism between 6-mercaptopurine a...

Conversion of 6-thioguanine to the nucleoside level by purine nucleoside phosphorylase of sarcoma 180 and sarcoma 180/TG ascites cells.

Tumor sublines sensitive (Sarcoma 180) and resistant (Sar coma 180/TG) to the cytotoxic action of 6-thioguanine (6TG) actively metabolize the 6-thiopurine to 6-thioguanosine (6TGuo). The conversion of 6TG to 6TGuo by intact cells was enhanced by D-ribose 1-phosphate but not by D-ribose 5phosphate. This corresponded to the finding that D-ribose 1phosphate supported the synthesis of 6TGuo from 6T...
